          Stem121:抗人類細(xì)胞質(zhì)蛋白的小鼠抗體

          1. Stem121是什么?

          · Stem121是小鼠來源的單克隆抗體,它特異性地識(shí)別人類細(xì)胞(特別是人類神經(jīng)細(xì)胞)細(xì)胞質(zhì)中的某個(gè)抗原。

          · Stem121在反應(yīng)屬性上有兩個(gè)主要特點(diǎn):

              1) 它所識(shí)別的抗原不在細(xì)胞核內(nèi),可以特異性的識(shí)別細(xì)胞質(zhì)內(nèi)的抗原;

              2)它與小鼠、大鼠、猴子細(xì)胞的細(xì)胞質(zhì)沒有反應(yīng)性。

          2. Stem121的應(yīng)用

          · 小鼠、大鼠、猴子是目前干細(xì)胞治療研究者常用的模式動(dòng)物。通常,研究者將人類的神經(jīng)干細(xì)胞注射進(jìn)入這些模式動(dòng)物的腦部,然后觀察這些細(xì)胞是否可以在動(dòng)物的腦部存活、生長(zhǎng)、分化、遷移。因此,必須可靠而且準(zhǔn)確地示蹤在動(dòng)物體內(nèi)的人類神經(jīng)干細(xì)胞。

          · Stem121抗體是可以對(duì)人類細(xì)胞細(xì)胞質(zhì)進(jìn)行標(biāo)記的抗體。

          · 對(duì)于輸入的人神經(jīng)干細(xì)胞,既標(biāo)記細(xì)胞核又標(biāo)記細(xì)胞質(zhì),意味著更加可靠、更加準(zhǔn)確。如下圖所示,人類的神經(jīng)干細(xì)胞被注射進(jìn)入大鼠的腦部,藍(lán)色標(biāo)記是某種識(shí)別人類細(xì)胞的細(xì)胞核的抗體,綠色標(biāo)記是識(shí)別人類細(xì)胞細(xì)胞質(zhì)的Stem121抗體,紅色標(biāo)記是識(shí)別人類細(xì)胞某種發(fā)炎癥狀的抗體。在A部分的左圖左下角有個(gè)箭頭指示的細(xì)胞,具備藍(lán)色與綠色標(biāo)記,這說明這個(gè)細(xì)胞確定是人類細(xì)胞而非大鼠細(xì)胞;右圖右下角的箭頭指示同一個(gè)細(xì)胞,沒有綠色標(biāo)記,說明這個(gè)人類細(xì)胞并沒有出現(xiàn)發(fā)炎癥狀。而且,如果實(shí)驗(yàn)的設(shè)計(jì)目的是觀察人類的神經(jīng)細(xì)胞的細(xì)胞質(zhì)能否進(jìn)入到人類或是動(dòng)物細(xì)胞的細(xì)胞核里面,那么就需要Stem121對(duì)人類細(xì)胞質(zhì)進(jìn)行標(biāo)記。
